Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014679.5(CEP57):c.589A>G (p.Asn197Asp), citing Ambry Variant Classification Scheme 2023: The p.N197D variant (also known as c.589A>G), located in coding exon 5 of the CEP57 gene, results from an A to G substitution at nucleotide position 589. The asparagine at codon 197 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
