NM_007294.4(BRCA1):c.3769GAG[1] (p.Glu1258del) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This variant deletes 3 basepairs in exon 10 in the BRCA1 gene, resulting in the in-frame deletion of a single amino acid, glutamic acid, at codon 1258 in the BRCA1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with BRCA1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
