Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.37C>T (p.Leu13Phe), citing Ambry Variant Classification Scheme 2023: The p.L13F variant (also known as c.37C>T), located in coding exon 1 of the SMAD6 gene, results from a C to T substitution at nucleotide position 37. The leucine at codon 13 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
