This variant lies in the ATP2A1 gene (transcript NM_004320.6) at the canonical splice donor site of the intron immediately after coding-DNA position 2980, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 21 of the ATP2A1 gene. While this variant is not anticipated to result in nonsense mediated decay, it likely alters RNA splicing and results in a disrupted protein product. This variant is present in population databases (rs753539984, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with ATP2A1-related conditions.
