Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020041.3(SLC2A9):c.463A>G (p.Met155Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 155 of the SLC2A9 protein (p.Met155Val). This variant is present in population databases (rs369512758, gnomAD 0.03%). This missense change has been observed in individual(s) with hypouricemia (PMID: 34572357). This variant is also known as p.Met126Val. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects SLC2A9 function (PMID: 34572357). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
