Uncertain significance for Hepatic veno-occlusive disease-immunodeficiency syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080424.4(SP110):c.145A>G (p.Met49Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 49 of the SP110 protein (p.Met49Val). This variant is present in population databases (rs766140257, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SP110-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
