Uncertain significance for Aortic valve disease 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005585.5(SMAD6):c.1416G>C (p.Trp472Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan, which is neutral and slightly polar, with cysteine, which is neutral and slightly polar, at codon 472 of the SMAD6 protein (p.Trp472Cys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SMAD6-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SMAD6 protein function with a positive predictive value of 80%.
