NM_000394.4(CRYAA):c.466G>A (p.Glu156Lys) was classified as Uncertain significance for Cataract 9 multiple types by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 156 of the CRYAA protein (p.Glu156Lys). This variant is present in population databases (rs76740365, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with CRYAA-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects CRYAA function (PMID: 36338667). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
