Uncertain significance for Ehlers-Danlos syndrome progeroid type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007255.3(B4GALT7):c.422G>A (p.Arg141Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 141 of the B4GALT7 protein (p.Arg141Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with B4GALT7-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt B4GALT7 protein function with a positive predictive value of 80%. This variant disrupts the p.Arg141 amino acid residue in B4GALT7. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 25533962, 26940150, 30914273).
