Submitter rationale: Variant summary: ALPL c.1471G>C (p.Gly491Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 241684 control chromosomes (gnomAD). c.1471G>C has been observed in individuals affected with Hypophosphatasia (Simon-Bouy_2008, Taillandier_2017). These data indicate that the variant may be associated with disease. A different variant resulting in the same amino acid consequence has been classified as pathogenic by our lab (c.1471G>A), supporting the pathogenicity of this variant. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
