NM_004360.5(CDH1):c.1002C>G (p.Asp334Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D334E variant (also known as c.1002C>G), located in coding exon 7 of the CDH1 gene, results from a C to G substitution at nucleotide position 1002. The aspartic acid at codon 334 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
