Uncertain significance for Progressive familial heart block type IB — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017636.4(TRPM4):c.1050_1050+19del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TRPM4 gene (transcript NM_017636.4) at coding-DNA position 1050 through 19 bases into the intron immediately after coding-DNA position 1050, deleting this region. Submitter rationale: This variant results in the deletion of part of exon 8 (c.1050_1050+19del) of the TRPM4 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TRPM4 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TRPM4-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
