Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003047.5(SLC9A1):c.859G>A (p.Gly287Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 287 of the SLC9A1 protein (p.Gly287Ser). This variant is present in population databases (rs145043085, gnomAD 0.09%). This variant has not been reported in the literature in individuals affected with SLC9A1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
