It does not change the encoded amino acid sequence of the RB1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with bilateral retinoblastoma (PMID: 30031154; internal data). Algorithms developed to predict the effect of variants on gene product structure and function are not available or were not evaluated for this variant. This variant is expected to affect transcription factor binding sites (SP1, ATF) in the RB1 promoter region and disruption of these binding sites affects RB1 transcription activity (PMID: 8049153, 8183566, 1881452, 33258708). A different variant within the promoter region (c.-198G>A) has been determined to be pathogenic (PMID: 1881452, 20447117, 10673998, Invitae). For these reasons, this variant has been classified as Pathogenic.
