Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.12:g.109465664C>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 5 of the MICAL1 protein (p.Ser5Asn). This variant also falls at the last nucleotide of exon 1, which is part of the consensus splice site for this exon. This variant is present in population databases (rs775871899, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with MICAL1-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
