This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 10718 through 5 bases into the intron immediately after coding-DNA position 10725, deleting this region. Submitter rationale: This variant results in the deletion of part of exon 75 (c.10718_10725+5del) of the RYR2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in RYR2 cause disease.
