Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000390.4(CHM):c.537A>C (p.Glu179Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 179 of the CHM protein (p.Glu179Asp). This variant is present in population databases (rs766404520, gnomAD 0.008%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with CHM-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
