Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.529C>G (p.Arg177Gly), citing Ambry Variant Classification Scheme 2023: The p.R177G variant (also known as c.529C>G), located in coding exon 3 of the RET gene, results from a C to G substitution at nucleotide position 529. The arginine at codon 177 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
