Uncertain significance for Combined deficiency of sialidase AND beta galactosidase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000308.4(CTSA):c.559C>G (p.Leu187Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 205 of the CTSA protein (p.Leu205Val). This variant is present in population databases (rs373498418, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with CTSA-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CTSA protein function.
