NM_000153.4(GALC):c.1851del (p.Ile616_Tyr617insTer) was classified as Pathogenic for Galactosylceramide beta-galactosidase deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GALC gene (transcript NM_000153.4) at coding-DNA position 1851, deleting one base. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 370366). This premature translational stop signal has been observed in individual(s) with Krabbe disease (PMID: 23319190). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr617*) in the GALC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GALC are known to be pathogenic (PMID: 7437911, 9272171, 16607461).
