Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a canonical 3' acceptor site. Three predict the variant creates a cryptic 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4.4e-05 in 251436 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in MCCC2 causing Methylcrotonyl-CoA Carboxylase Deficiency (4.4e-05 vs 0.0042), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.384-19T>A in individuals affected with Methylcrotonyl-CoA Carboxylase Deficiency and no experimental evidence demonstrating its impact on protein function have been reported.
