Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.8345A>G (p.His2782Arg), citing Ambry Variant Classification Scheme 2023: The p.H2782R variant (also known as c.8345A>G), located in coding exon 64 of the FBN2 gene, results from an A to G substitution at nucleotide position 8345. The histidine at codon 2782 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
