This variant lies in the MYH11 gene (transcript NM_002474.3) at coding-DNA position 3858, where G is replaced by C; at the protein level this means replaces glutamine at residue 1286 with histidine — a missense variant. Submitter rationale: This sequence change replaces glutamine, which is neutral and polar, with histidine, which is basic and polar, at codon 1293 of the MYH11 protein (p.Gln1293His). This variant also falls at the last nucleotide of exon 29, which is part of the consensus splice site for this exon. This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with MYH11-related conditions.
