Pathogenic for Glycogen storage disease, type II — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000152.5(GAA):c.1194+2T>C, citing Genomenon Sequence Variant Interpretation Standards - Updated: GAA c.1194+2T>C is a canonical splice variant affecting the donor splice site of intron 7. It is predicted to affect mRNA splicing, leading to a deleterious effect on the GAA protein. This variant has been observed in at least one proband with a GAA-related disorder (PMID:36137614;34353347;31953985;31193175;22658377). At least one splicing study has demonstrated that this variant results in aberrant splicing (PMID:18458862). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GAA c.1194+2T>C as a pathogenic variant.
