While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 36 amino acid(s) of the FANCG protein. This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with FANCG-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant disrupts the C-terminus of the FANCG protein. Other variant(s) that disrupt this region (p.Leu591Argfs*3) have been observed in individuals with FANCG-related conditions (Invitae).
