Uncertain significance for Gastrointestinal stromal tumor — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006206.6(PDGFRA):c.2324-18_2335del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PDGFRA gene (transcript NM_006206.6) at 18 bases into the intron immediately before coding-DNA position 2324 through coding-DNA position 2335, deleting this region. Submitter rationale: This variant results in the deletion of exon 17 (c.2324-18_2335del) of the PDGFRA gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in PDGFRA cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PDGFRA-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
