Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012478.4(WBP2):c.239G>A (p.Cys80Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 80 of the WBP2 protein (p.Cys80Tyr). This variant is present in population databases (rs771731567, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with WBP2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt WBP2 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
