Uncertain significance for Developmental and epileptic encephalopathy, 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001353921.2(ARHGEF9):c.23C>T (p.Ser8Leu), citing Invitae Variant Classification Sherloc (09022015): The ARHGEF9 gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_001173479.1, and corresponds to NM_015185.2:c.-30811C>T in the primary transcript. This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 8 of the ARHGEF9 protein (p.Ser8Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ARHGEF9-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
