Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_016222.4(DDX41):c.1010T>A (p.Ile337Asn), citing Ambry Variant Classification Scheme 2023: The p.I337N variant (also known as c.1010T>A), located in coding exon 10 of the DDX41 gene, results from a T to A substitution at nucleotide position 1010. The isoleucine at codon 337 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
