Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033100.4(CDHR1):c.2269A>C (p.Lys757Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with glutamine, which is neutral and polar, at codon 757 of the CDHR1 protein (p.Lys757Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of CDHR1-related conditions (Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CDHR1 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
