Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004972.4(JAK2):c.3376A>G (p.Ile1126Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1126 of the JAK2 protein (p.Ile1126Val). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with JAK2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt JAK2 protein function with a negative predictive value of 80%.
