Uncertain significance for Treacher Collins syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001371623.1(TCOF1):c.4036C>A (p.Arg1346Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 1345 of the TCOF1 protein (p.Arg1345Ser). This variant is present in population databases (rs781234452, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with TCOF1-related conditions. This missense change has been observed in at least one individual who was not affected with TCOF1-related conditions (Invitae). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
