Uncertain significance for 3-methylcrotonyl-CoA carboxylase 1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020166.5(MCCC1):c.1592A>G (p.His531Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 531 of the MCCC1 protein (p.His531Arg). This variant is present in population databases (rs749202264, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with MCCC1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
