Pathogenic for Diamond-Blackfan anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001022.4(RPS19):c.407dup (p.Gln137fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the RPS19 gene (p.Gln137Thrfs*17). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 9 amino acid(s) of the RPS19 protein and extend the protein by 7 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This frameshift has been observed in individual(s) with clinical features of Diamond-Blackfan anemia (internal data). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
