NM_016599.5(MYOZ2):c.251G>A (p.Ser84Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S84N variant (also known as c.251G>A), located in coding exon 3 of the MYOZ2 gene, results from a G to A substitution at nucleotide position 251. The serine at codon 84 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
