Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.2200T>G (p.Tyr734Asp), citing Ambry Variant Classification Scheme 2023: The p.Y734D variant (also known as c.2200T>G), located in coding exon 12 of the ALK gene, results from a T to G substitution at nucleotide position 2200. The tyrosine at codon 734 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
