Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014283.5(SUCO):c.3556A>G (p.Ser1186Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 1186 of the SUCO protein (p.Ser1186Gly). This variant is present in population databases (rs765663781, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with SUCO-related conditions. ClinVar contains an entry for this variant (Variation ID: 3671987). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
