Likely pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024675.4(PALB2):c.2515-17_2522del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exon 6 of the PALB2 gene. RNA analysis indicates that this variant induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PALB2-related conditions. ClinVar contains an entry for this variant (Variation ID: 3671507). Algorithms developed to predict the effect of variants on gene product structure and function are not available or were not evaluated for this variant. Experimental studies have shown that this variant affects PALB2 function (PMID: 26990772, 30890586).
