Uncertain significance for Charcot-Marie-Tooth disease axonal type 2F — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001540.5(HSPB1):c.365-14_365-7del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the HSPB1 gene (transcript NM_001540.5) at 14 bases into the intron immediately before coding-DNA position 365 through 7 bases into the intron immediately before coding-DNA position 365, deleting this region. Submitter rationale: This sequence change falls in intron 1 of the HSPB1 gene. It does not directly change the encoded amino acid sequence of the HSPB1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with HSPB1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
