Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001372.4(DNAH9):c.308T>A (p.Phe103Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with tyrosine, which is neutral and polar, at codon 103 of the DNAH9 protein (p.Phe103Tyr). This variant is present in population databases (rs373784591, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DNAH9-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt DNAH9 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
