Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130004.2(ACTN1):c.898C>T (p.Arg300Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 300 of the ACTN1 protein (p.Arg300Trp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with isolated nonsyndromic thrombocytopenia (PMID: 32757236). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ACTN1 protein function with a positive predictive value of 80%.
