Pathogenic for Argininosuccinate lyase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000048.4(ASL):c.524+1G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ASL gene (transcript NM_000048.4) at the canonical splice donor site of the intron immediately after coding-DNA position 524, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 7 of the ASL gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with argininosuccinate lyase deficiency (PMID: 17326097). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Studies have shown that disruption of this splice site results in skipping of skipping of exon 7 (also known as exon 6), but is expected to preserve the integrity of the reading-frame (PMID: 17326097). For these reasons, this variant has been classified as Pathogenic.
