Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_003995.4(NPR2):c.2833C>T (p.Arg945Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: NPR2 c.2833C>T (p.Arg945Cys) results in a non-conservative amino acid change located in the Adenylyl- / guanylyl cyclase, catalytic domain (IPR001054) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 251468 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2833C>T in individuals affected with NPR2-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported.
