Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003737.4(DCHS1):c.286G>T (p.Glu96Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu96*) in the DCHS1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DCHS1 are known to be pathogenic (PMID: 24056717, 26258302). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DCHS1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
