NM_001042492.3(NF1):c.2252-2A>T was classified as Pathogenic for Neurofibromatosis, type 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NF1 gene (transcript NM_001042492.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2252, where A is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 18 of the NF1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with neurofibromatosis, type 1 (PMID: 12872266). Studies have shown that disruption of this splice site results in skipping of exon 19, and produces a non-functional protein and/or introduces a premature termination codon (internal data). For these reasons, this variant has been classified as Pathogenic.
