Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003079.5(SMARCE1):c.937C>G (p.Arg313Gly), citing Ambry Variant Classification Scheme 2023: The p.R313G variant (also known as c.937C>G), located in coding exon 9 of the SMARCE1 gene, results from a C to G substitution at nucleotide position 937. The arginine at codon 313 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
