Uncertain significance for Charcot-Marie-Tooth disease type 2B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004637.6(RAB7A):c.612C>G (p.Ser204Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 204 of the RAB7A protein (p.Ser204Arg). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAB7A-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
