Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001190274.2(FBXO11):c.717+5G>C, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FBXO11 gene (transcript NM_001190274.2) at 5 bases into the intron immediately after coding-DNA position 717, where G is replaced by C. Submitter rationale: This sequence change falls in intron 5 of the FBXO11 gene. It does not directly change the encoded amino acid sequence of the FBXO11 protein. It affects a nucleotide within the consensus splice site. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with FBXO11-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
