The next in-frame methionine is located at codon 20. This variant is not present in population databases (gnomAD no frequency). Disruption of the initiator codon has been observed in individual(s) with PLN-related conditions (PMID: 30242101). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant disrupts a region of the PLN protein in which other variant(s) (p.Arg9Cys) have been determined to be pathogenic (PMID: 12610310, 21282613, 22427649, 22707725, 25593317).
