Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.4919T>A (p.Val1640Glu), citing Ambry Variant Classification Scheme 2023: The p.V1640E variant (also known as c.4919T>A), located in coding exon 40 of the CACNA1C gene, results from a T to A substitution at nucleotide position 4919. The valine at codon 1640 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
